چکیده

The fermentation of deoxyribose in Escherichia coli (1, 2) and in Lactobacillus plantarum (3, 4) appears to involve 2-deoxyribose 5-phosphate as an intermediate. Racker (2) has described an enzyme, deoxyribose phosphate aldolase, which catal.yzes the reversible cleavage of deoxyribose 5-phosphate to acetaldehyde and glyceraldehyde 3-phosphate. The enzyme occurs in E. coli and in animal tissues (2), where its function is not clearly established, and in deoxyribose-grown cells of L. plantarum, where it plays a major role in the fermentation pathway (3). The present paper is a report of studies on the induction, purification, and kinetic properties of deoxyribose phosphate aldolase of L. plantarum, together with a method for the synthesis and isolation of 2-deoxyribose 5-phosphate.

Deoxyribose Phosphate Aldolase from Rat Liver.

The activity and properties of deoxyribose phosphate aldolase from Escherichia coli and Lactobacillus plantarum have been described in detail by Racker (1) and Pricer and Horecker (2). The enzyme from E. coli was shown to be active at pH values between 6.0 and 8.0 in tris(hydroxymethyl)aminomethane and phosphate buffers without exhibiting a sharp optimum (1). The enzyme from L. plunturum was sh...

2-deoxyribose gene-enzyme complex in Salmonella typhimurium. I. Isolation and enzymatic characterization of 2-deoxyribose-negative mutants.

Salmonella typhimurium was found to utilize 2-deoxyribose as a sole carbon and energy source. Cells grown in the presence of deoxyribose contained increased levels of deoxyribose kinase, thymidine phosphorylase, and two forms of deoxyribose-5-phosphate aldolase (DR5P aldolase). One form of DR5P aldolase was induced by deoxyribose and coordinately regulated with deoxyribose kinase. The second fo...

2-Deoxyribose gene-enzyme complex in Salmonella typhimurium: regulation of phosphodeoxyribomutase.

Phosphodeoxyribomutase, the enzyme which catalyzes the interconversion of 2-deoxyribose-1-phosphate to 2-deoxyribose-5-phosphate, has been partially purified from Salmonella typhimurium. The enzyme had an absolute requirement for manganese ion and was stimulated by glucose-1, 6-diphosphate. Phosphodeoxyribomutase was induced by deoxyribose-5-phosphate and was coordinately regulated with the enz...

An apparent lack of stereospecificity in the reaction catalysed by deoxyribose 5-phosphate aldolase due to methyl-group rotation and enolization before product release.

In the reaction catalysed by deoxyribose 5-phosphate aldolase (2-deoxy-D-ribose 5-phosphate acetaldehyde-lyase, EC 4.1.2.4) from Salmonella typhimurium, almost complete equilibration of the methyl-group protons of the product, acetaldehyde, occurs before its release from the enzyme surface. This phenomenon does not allow the stereo-chemical course of the reaction to be determined by using hydro...

Acrolein, an irreversible active-site-directed inhibitor of deoxyribose 5-phosphate aldolase?

The enzyme deoxyribose 5-phosphate aldolase was irreversibly inactivated by the substrate analogue acrolein with a pseudo-first-order rate constant of 0.324 min-1 and a Ki (apparent) of 2.7 x 10(-4) m. No inactivation was observed after prolonged incubation with the epoxide analogues glycidol phosphate and glycidaldehyde. It is suggested that the acrolein is first activated by forming a Schiff ...
